Do you have to rack the slide every time you use it ?

if your pistol is double action then no. if your trigger operates as single action then yes (but you usually only have to pull the slide back 1/4 - 1/2 inch or so to reset and not all the way back so as to let the slide stop grab it (if you have an empty mag in it) you can also remove the follower from a mag and use that for practice if you have difficulty racking the slide back only part way. I am an instructor and use a taurus g3c for laser trainers like this because it is a semi-auto with a "double-action" trigger as a striker fire

Can I manually set this laser on and off while in a barrel of a gun to calibrate with this laser a picatinny rail laser/flashlight gun attachment?

When you push the On/Off button, the Laser briefly flashes a beam. What you need is a Boresight. They make that, too, and it works great and is fairly cheap. I have one. I know.

Can it be used with Rugger lcp 380?
