What cleaner should I use for this floor?

You can use pretty much any mild floor cleaner. I would recommend applying it with a microfiber mop and not just dumping it directly on the floor. The Turbo Mop has worked well for me.

Do 20 come in the pack? I have a small 4x4 area to cover, so wondering if this is just for ONE tiny 12 inch square, or is this an entire pack?

do the tiles create a waterproof floor?

I would say no. The adhesive they use is great, but doesn't make it waterproof.
