What is the maximum load these wheels can take?

I don't know the specific weight the replacement wheel is rated for. One would have to contact the manufacturer for that. However, since the the wheel is a replacement wheel for luggage that is the size that is for aircraft luggage compartment ( not carry on board ) I would assume that the wheels meet the Airline Luggage Weight requirement for large luggage.

Will these fit Samsonite wheeled briefcase? Do you supply instructions?

I did not receive instructions, but the product worked great in my case. I don't know if it will work for samsonite wheeled briefcase.

You will absolutely need to measure the outer diameter of the wheels on your case. If you can easily remove the existing wheels, pull one and measure the axle diameter. While you're at it, measure the width of the wheel at the axle bearings. The kit comes with two different length axles, so that more or less takes the worry out of that part of the equation.
