Will this valve fit a Quest queen sized air mattress?

Sorry, I can't help you here. I bought the air valve(s) to replace those lost on a inflatable 3 person boat, which worked perfectly, I might add.

Will it work with the river run inflatable tubes?

Yes. it will work with the Intex River Run tube as well as many other Intex devices such as the Seahawk II

How do you inflate with the Boston valve.

I use a normal air pump like this: http://www.amazon.com/gp/product/B000FE5PBA?psc=1&redirect=true&ref_=oh_aui_detailpage_o09_s00Unscrew the top portion of the boston valve and choose the valve that fits into it and just let it pump.

what is the thread diameter?

From what I can tell when I ordered thread diameter is 7/8" and 15/16". The large one fit my airhead tube
