Can a large need and yarn fit through the holes

Regular worsted weight #4 yarn will fit, but you will need to use a small needle. A large eye needle will not go through.

Are they washing machine friendly?

We used these buttons on costumes for Seven Brides for Seven Brothers. They were on the bright colorful shirts the brothers wear at the Barn raising (for the movie). They had to endure two weeks worth of dress rehearsals and eight performances. They were washed after every use because our boys were quite sweaty with all that dancing. The buttons were perfectly attached after each wash. I was very pleased.

Would 2mm leather cord fit through the holes?

No, you would have to drill the holes larger.
