Q: Can this be thrown in the washing machine?

Yes...gentle cold water cycle... air dry.

Q: Is the strap adjustable?

No. It's just a strap; the bag is sewn as two halves, basically, so everything is sort of one piece. I am 5'8", and it hits me at right about the hip.

Q: Does it fit a binder?

It's a big bag,I can't fit my laptop (It's really big) but a binder would fit but keep in mind that all the pressure is on that one shoulder (Don't be worried about the stitching)

Q: Can it hold clothes, makeup bag, water bottle, etc? And Is there any sort of pocket?

The medium can definitely hold a make up bag and water bottle, maybe a piece of lightweight clothing. I wouldn't use it like a small duffle bag though. I had my iPad in it, Wallet, small personal items, souvenirs etc. For clothing I would suggest the large size. I'm very happy with the medium. Yes, it does have a small pocket inside.
